Exelixis is a leading genomics-based drug discovery company focused on the discovery and development of innovative new drugs for cancer and other major human diseases. Through its integrated biological platform and expertise in comparative genomics and model system genetics, Exelixis is able to find novel, proprietary drug targets that would be difficult or impossible to uncover using other approaches. Internally, Exelixis has built an aggressive high-throughput screening and chemistry facility in order to accelerate these targets through the drug discovery process. The remarkable evolutionary conservation of biochemical pathways strongly supports the use of simple model systems, such as fruit flies, nematode worms, zebrafish, and mice to identify key members of critical disease pathways that can then be targeted for drug discovery. Specifically in cancer, Exelixis exploits the underlying "genetic liabilities" of tumor cells to identify biologically relevant drug targets, which, when modulated by a drug, specifically kill those cells while leaving normal cells unharmed. The company has discovered for its own development efforts and delivered to collaborators a substantial number of highly interesting cancer targets. Many of these are being pursued internally, while others are being shared with Bristol-Myers Squibb and utilized as monoclonal antibody cancer drug targets in a collaboration with Protein Design Labs. In its pioneering collaboration with Bristol-Myers Squibb, Exelixis is sharing these targets to create a potentially new generation of small molecule cancer therapeutics. Molecules and antibodies developed against these targets may selectively kill cancer cells while leaving normal cells unharmed, and may provide alternatives to current cancer therapies.
